Using tea stem slicing and dyeing technologies, the age of tea plants was accurately determined based on the digital images. Under ordinary light and fluorescence of bio-digital microscope, the untreated sections of tea stem, saffron-dyed sections and acridine orange-dyed sections of tea stem were compared and analyzed. The result shows that acridine orange dyeing was the best to determine the age of tea plants. The acridine orange dyeing method combined with digital image analysis had the characteristics of fast, accurate, simple and low cost. In conclusion, the technology also had a scientific reference significance for the identification of annual rings of living large and ancient tea plants.
