Bio-efficacy of Tea Extract on Mouse B16 Melanoma Cell in Vitro and Its Mechanism

Abstract: The Bio-efficacy and mechanism of tea extract on mouse B16 melanoma cell in vitro were studied in this paper. Cells were treated with different concentration of TF1, EGCG and TF40, then the cell morphology was observed. The tyrosinase activity was determined by MTT method. The synthesis of melanin was determined using DOPA as substrate, and proliferation rate of B16 cells was determined by colorimetry, then analyzing the effects of drugs to these indexes. The results showed that the cell morphology was changed by the treatment of drugs. The tyrosinase activity, the synthesis of melanin and proliferation rate of B16 cells were all negatively related to the drugs concentration. Tea extracts and Vc can cause the inhibition of the synthesis of melanin, and resulted to white skin. The whiting effect of tea extracts is better than that of Vc.
