水培茶树累积铅在细胞内分布的TEM-EDS超微定位表征

Characterization of Pb after Accumulated and Localized in Ultra-structure Cells of Hydroponic Tea Plant

摘要: 应用水培茶树模式,采用透射电子显微镜(Transmission Electron Microscope,TEM)和X射线能谱仪(Energy Dispersive Spectrometer,EDS)相结合的手段,对铅盐水培茶树根、茎、叶细胞内Pb2+的累积部位与分布水平进行观测与分析。结果表明:茶树根细胞中的铅均匀地分布于细胞壁上,且数量最多,质膜、细胞质和液泡膜上的铅较少,细胞间隙等部位的铅最少;嫩茎细胞壁中的铅颗粒小、数量多,质膜、液泡膜及叶绿体上数量较少;叶片细胞中的铅则主要分布在质膜和液泡膜上,细胞壁中的铅较少。

Abstract: This study applied lead salts hydroponic tea plant, observed and analyzed Pb accumulation and ultra-structure localization in the cells of root, stem, leaves by the TEM-EDS method. The results showed that most of Pb evenly distributed on the cell wall of root cells, only few Pb distributed on the plasma, cytoplasm, vacuole membrane and intercellular space. While, Pb on cell wall of stem were numerals and small, and a little on plasma, vacuole membrane and chloroplast. Pb in leaves were mainly on plasma and vacuole membrane, there were only a little on the cell wall of leaves.
