高效液相色谱-蒸发光散射检测器测定茶叶中茶氨酸

Determination of Theanine in Tea by Reversed-Phase High Performance Liquid Chromatography-Evaporative Light-Scattering Detector

摘要: 建立未衍生化高效液相色谱-蒸发光散射检测器测定茶叶中茶氨酸。采用Polaris-C18(250βmm×4.6βmm)色谱柱,以含0.1%(体积分数)三氟乙酸的水和0.1%(体积分数)三氟乙酸的乙腈作梯度洗脱,流速0.5βml/min,蒸发光散射检测器为ELSD-2000。茶氨酸浓度在0.062~0.450βmg/ml内,其峰面积的常用对数与进样量的常用对数呈良好的线性关系,平均回收率为97.5%。本方法具有精确、灵敏等特点。

Abstract: High performance liquid chromatography (HPLC) coupled with evaporative light scattering detector(ELSD-2000)was used for the determination of theanine in tea without derivatization. A Polaris-C18 column (250βmm×4.6βmm) was used with the mobile phase of 0.1%trifluoroacetic acid water solution and 0.1% trifluoroacetic acid acetonitrile solution .The flow rate was at 0.5βml/min. The linear response ranged from 0.062~0.450βmg/mL for theanine. The recovery was 97.5%. The method was evaluated as accurate and simple.
